      目的:探讨MMP-1,MMP-13 在慢性睡眠限制引起大鼠髁突软骨结构变化中的表达变化及作用。方法:180 只雄性Wistar大 鼠随机分为3 组(n=60):慢性睡眠限制组(CSR)、大平台组(LC)、笼养组(CON)。每组根据试验时间不同分别分为3 个亚组 (n=20):7 天(7D)、14 天(14D)、21 天(21D)组。参考改良多平台法(MMPM)建立大鼠的慢性睡眠限制模型。通过HE染色观察大 鼠髁突软骨的结构变化。通过免疫组化和实时定量PCR 分别检测MMP-1 和MMP-13 的蛋白水平及mRNA水平的表达变化。结 果:HE 染色和扫描电镜结果显示,CSR 组的大鼠髁突软骨出现了病理性的改变。与CON 和LC 组比较,CSR 组MMP-1 和 MMP-13 的mRNA转录和蛋白表达水平明显升高(P<0.05)。结论:慢性睡眠限制能够引起大鼠颞下颌关节髁突软骨的病理性变 化。MMP-1 和MMP-13 的表达水平的变化可能在大鼠髁突软骨病理性改变中起关键作用。

      Objective:To explore the changes and possible roles of MMP-1 and MMP-13 in the condylar cartilage of rats subjected to chronic sleep restriction (CSR).Methods:One hundred and eighty male Wistar rats were randomly divided into three groups (n = 60 in each group): chronic sleep restriction group (CSR), large-platformcontrol group (LC) and cage control group (CON). Each group was divided into three subgroups (n = 20 in each subgroup) of three different time points (7, 14 and 21 days), respectively. The modified multiple platform method was used to induce chronic sleep restriction. The condylar cartilage tissue histology was studied by staining with haematoxylin and eosin (HE). The expression of MMP-1 and MMP-13 proteins and mRNAs was detected by immunohistochemistry and real-time polymerase chain reaction (RT-PCR), respectively.Results:HE staining showed pathological alterations in the condylar cartilage following CSR. Compared with CON and LC groups, the mRNA and protein expression levels of MMP-1 and MMP-13 were upregulated after CSR (P<0.05).Conclusion:These data suggest that chronic sleep restriction may induce pathological alterations in the condylar cartilage of rats. Alterations in MMP-1 and MMP-13 may be play an important role in the mechanismby which CSR influences the condylar cartilage.
