目的:探讨杀伤细胞对人Lewis肺癌细胞增殖的抑制作用。方法:选择成年健康雄性ICR 小鼠46只,随机选取10 只作为正
常对照组,其余36 只进行模型复制,然后将造模成功的30 只小鼠随机分为实验组与模型组,每组15 只。实验组予以尾静脉注射
杀伤细胞1× 107个/0.2 mL,1 次/d,模型组与对照组予尾静脉注射生理盐水。ELISA 方法测定TNF-alpha、IL-2 和IL-4 的含量,采用
流式细胞仪检测凋亡率,四氮唑蓝(MTT)比色法检测癌细胞抑制率。结果:①实验组小鼠右腋下瘤体重量明显低于模型组,差异
有统计学意义(P<0.01);②透射电镜下观察实验组人Lewis 肺癌细胞出现大量凋亡,而模型组人Lewis 肺癌细胞几乎未凋亡,与
模型组比较,实验组细胞凋亡较多,差异具有统计学意义(P<0.05);③实验组小鼠脾上清液中IL-2、TNF-alpha和IFN-gamma含量明显高
于模型组和正常对照组小鼠,差异有统计学意义(P<0.01)。结论:杀伤细胞能够通过活化淋巴细胞、分泌细胞因子来抑制体内、体
外人Lewis 肺癌细胞的增殖和生长,对临床具有指导意义。 |

Objective:To investigate inhibitory role of Lewis killer cells against the proliferation and growth of lung cancer cell.Methods:46 male ICR mice aged 8 weeks, weighing 22-25 g were selected. 10 rats were randomly selected as normal control group, and
the rest 30 rats were established the model, and when the model was successfully established, the rats were divided into two groups. The
experimental group was intravenous injected of killer cells to 1× 107 /0.2 mL, once a day, and the rats of the model group and the control
group received intravenous injection of saline. The level of TNF-alpha, IL-2 and IL-4 was detected by ELISA, and the apoptosis was detected
by flow cytometry, and the inhibition rate of cancer cells was detected by MTT.Results:Compared with the control group, the right
axillary tumor weight in experimental group was lower, and the difference was statistically significant (P<0.01). The Lewis human lung
cancer cell presented a large amount of apoptosis in the experimental group, but almost no apoptosis was detectable in the model group
with statistical significance between two groups (P<0.05). The spleen supernatant IL-2, TNF- alpha and IFN- gamma were significantly
higher in the experimental group than that of the model group and normal control group, and the difference was statistically significant
(P<0.01).Conclusion:Killer cells can inhibit the proliferation and growth of Lewis lung cancer cell through activating the lymphocytes
and secreting the cytokines. |
