      目的:探讨MIF和CerbB-2的表达与子宫内膜癌的关系,进一步探讨其表达与子宫内膜癌的发生、发展的关系,为早期诊断 子宫内膜癌,判断预后和寻找子宫内膜癌的新的治疗方法奠定基础。方法:应用实时荧光定量RT-PCR 和免疫组织化学技术检测 正常子宫内膜、不典型增生和子宫内膜癌组织中MIF mRNA、CerbB-2 mRNA和MIF蛋白、CerbB-2 蛋白的表达,其中免疫组化中 子宫内膜癌组根据临床病理特征,如年龄,临床分期,组织学分级,肌层浸润深度和有无淋巴结转移进行分组。结果:正常子宫内 膜、不典型增生内膜和子宫内膜癌组织中都有MIF和CerbB-2 的表达,子宫内膜癌中MIF mRNA和CerbB-2 mRNA 扩增含量显 示高水平,其阳性表达率随着病变程度的加重而逐渐升高,差异具有统计学意义(P=0.037);MIF主要表达于子宫内膜腺上皮的细 胞质中,少量表达于细胞质,在阳性表达的子宫内膜癌组织中,蛋白含量在I 期(P=0.033)、G1 期(P=0.034)、无淋巴结转移的组织 中水平较高(P=0.041),差异具有统计学意义。CerbB-2 蛋白主要表达在子宫内膜的细胞质,少量在细胞膜中表达,阳性表达率子 宫内膜癌>不典型增生>正常子宫内膜,差异具有统计学意义(P=0.013);CerbB-2 蛋白在III-IV(P=0.009)、G2-3 期(P=0.033),有 淋巴结转移(P=0.018)的子宫内膜癌中表达含量较高,与年龄和肌层浸润深度无关。结论:MIF和CerbB-2 在正常子宫内膜、不典 型增生和子宫内膜癌中都有表达,提示着MIF 和CerbB-2与子宫内膜癌的发生和发展有关,MIF 在子宫内膜癌的早期表达含量 较高,可以作为子宫内膜癌的早期诊断指标,CerbB-2在子宫内膜癌的晚期表达含量较高,可以促进肿瘤的侵袭转移,可作为判断 疾病的严重程度和预后的指标之一。

      Objective:To explore the expressions of MIF and CerbB-2 in endometrial cancer, and the relationship between their expression and the occurrence and development of endometrial carcinoma.Methods:15 cases of normal endometrium, 20 cases of atypical hyperplasia and 50 cases of endometrial carcinoma were collected from the fourth Affiliated Hospital of Harbin Medical University fromJanuary 2013 to September 2014. The expression of MIF and CerbB-2 in transcript and protein level was detected by real-time fluorescent quantitative and immunohistochemistry in normal endometrium, atypical hyperplasia and endometrial carcinoma. The cases of endometrial carcinoma group according to the clinical and pathological characteristics, such as age, clinical stage, histological grade, depth of myometrial invasion and status of lymph node metastasis were further subgrouped.Results:MIF and CerbB-2 was expressed in normal endometrium, atypical hyperplasia endometriumand endometrial carcinoma, MIF mRNA and CerbB-2 mRNA amplification content dis- played a high level in endometrial carcinoma, and the positive expressing rate was increased along with the severity degree of the disease, and the difference was statistically significant (P=0.034). MIF protein was expressed in endometrial cytosol, and little in nuclear The positive expression of MIF in endometrial carcinoma had higher level in stage I(P=0.033), stage G1(P=0.034), no lymph node metastasis tissues (P=0.041), and the difference was statistically significant. The CerbB-2 protein was expressed in the cytoplasm of endometrium, little in the membrane. The expression of CerbB-2 protein was higher in III-IV, G2-3 phase and lymph nodes metastasis, and its expression had no relationship with clinical stage, depth of myometrial invasion (P>0.05).Conclusion:MIF and CerbB-2 were expressed in normal endometrium, atypical hyperplasia and endometrial carcinoma, suggesting that MIF and CerbB-2 are correlated with the occurrence and the development of the endometrial cancer. The higher expression levels of MIF are in the early of endometrial cancer, which can be used for the early diagnosis of endometrial cancer, while CerbB-2 has the higher expression level in the late of endometrial cancer, and it can be an indicator of the severity of the disease and prognosis.
