      目的:观察和分析大鼠睾丸局部短暂热应激对HSP70、HSP90、HSP105 mRNA表达的影响。方法:Wistar 雄性大鼠随机分为 5 组:正常对照(N)组、热应激0 天(H0)组、热应激5 天(H5)组、热应激10 天(H10)组、热应激15 天(H15)组。N 组睾丸局部22℃水浴 20 min,其余各组均睾丸局部43℃水浴20 min。采用HE染色观察组织形态学变化,采用Real Time PCR 的方法检测HSP70、 HSP90、HSP105 mRNA的表达量。结果:HE 染色镜下观察结果显示:与N 组相比,H5组部分曲细精管萎缩,生精细胞明显消失, H10 组、H15 组大部分曲细精管萎缩,生精细胞大量消失。HE染色形态计量结果显示:与N 组相比,H5 组、H10 组、H15 组睾丸实 质体积比明显减小(p<0.05),睾丸间质体积比明显增加(P<0.05)。RT-PCR 结果显示:与N 组相比,H0 组HSP70 mRNA 的表达 H0 组明显增高(p< 0.05),H5 组、H10 组、H15 组HSP90 mRNA的表达明显降低(P< 0.05),H5 组HSP105 mRNA 的表达明显降 低(P< 0.05)。结论:HSP70、HSP90、HSP105 mRNA的表达在热应激后都会发生变化,变化的具体情况不尽相同,推测它们热应激 后在生殖细胞凋亡过程中发挥不同的作用有关,并且和组织的损伤有密切的联系。

      Objective:To observe and analyze the influence of brief heat stress on testical on the expression of HSP70, HSP90, HSP105 mRNA in rat testis.Methods:Male Wistar rats were randomly divided into normal control (N) group, heat stress 0 days (H0) group, heat stress 5 days (H5) group, heat stress 10 days (H10) group, heat stress 15 days (H15) group.N group testis werebathed in water for 20min at 22℃, and the rest were bathed in water for 20 min at 43℃ . Then morphological changes of testical tissues were observed by using HE staining and level of HSP70, HSP90, and HSP105 mRNA was detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR).Results:Microscopy with HE staining showed atrophy of some seminiferous tubules and significant disappearance of spermatogenic cells in H5 group, and atrophy of most seminiferous tubules and remarkable disappearance of spermatogenic cells in H10 and H15 group compared with N group. Morphometry with HE staining presented a significant decrease of testicular parenchyma volume ratio (P<0.05) and a significant increase of Leydig volume ratio (P<0.05) in H5, H10 and H15 group compared with N group. The results of RT-PCR demonstrated that compared with N group, the expression of HSP70 mRNA was Up-regulated significantly (P<0.05) in H0 group, that HSP90 mRNA expression wasdown-regulated significantly (P<0.05) in H5 group, H10 group and H15 group, and that HSP105 mRNA expression was down-regulated significantly (P<0.05) in H5 group.Conclusion:After the transient heat stress in rat testis, a relatively differential expression of HSPs occurred, which could suggest that HSPs play a different role in the process of heat stress-induced germcell apoptosis and that HSPs is closely related to tissue injury.
