Objective:To investigate the cytotoxicity of vancomycin (Van)/calcium phosphate cement (CPC) composite materials
on rat bone marrow mesenchymal stem cells (BMSCs).Methods:Primary BMSCs were isolated, cultured and identified. The cells were
incubated with different concentrations (containing 0%, 1%, 5%, 10% Van) of Van/CPC composite materials. The cell viability of
BMSCs was measured by CCK-8 method, and the cell apoptosis was detected by TUNEL method and the cell morphology was
observed-by scanning electron microscope (SEM).Results:5% and 10% Van/CPC composite materials could significantly inhibit the
proliferation of BMSCs (P<0.05), and increase apoptosis (P<0.05) and lead to higher cytotoxicity.1%Van/CPC exerted no obvious effect
on the BMSCs compared to the blank control.Conclusion:Low concentration of Van/CPC had no cytotoxicity on rat and BMSCs
performed good biocompatibility. |
