目的:研究小干扰RNA(short interfering RNA,siRNA)沉默beta-catenin 基因对胃癌AGS 细胞人端粒酶逆转录酶(human
telomerase reverse transcriptase,hTERT)的影响。方法:用Lipofectamine 2000 将siRNA-beta-catenin(实验组)及siRNA-control(阴性
对照组)序列转入细胞AGS中,同时设置正常AGS 组(空白对照组),运用细胞计数法检测三组细胞增殖能力的变化,同时使用
western blot检测三组细胞beta-catenin、hTERT 的表达。结果:转染72 h后,siRNA-beta-catenin组细胞增值能力显著低于siRNA-control
组和正常AGS 组(P< 0.05),siRNA-beta-catenin 组细胞beta-catenin、hTERT表达显著低于siRNA-control 组和正常AGS 组(P< 0.05)。
结论:AGS 细胞中Wnt/beta-catenin 信号通路可能参与调控hTERT基因转录。 |

Objective:To research the influence of beta-catenin silencing by short interfering RNA (siRNA) on human telomerase
reverse transcriptase (hTERT) of AGS cells, and explore its mechanism and relationships.Methods:siRNA-beta-catenin (experimental
group) and siRNA-control (negative control group) sequences were transfected into AGS cells by Lipofectamine 2000, and set the normal
AGS cells group (blank group) at the same time. The three groups of cells proliferation ability were detected by counting method, while
the expression of hTERT and beta-catenin were determined by Western blot.Results:72h after transfection, the proliferation ability of
siRNA-beta-catenin group were significantly lower than the siRNA-control group and normal AGS group (P <0.05), and the expression of
beta-catenin and hTERT were significantly lower than siRNA-control group and normal group (P<0.05).Conclusion:Wnt/beta-catenin
signaling pathway may be involved in the regulation of hTERT gene transcription. |
