滋mol/L 和 150 滋mol/L 过氧化氢处理组中细胞存活能力逐渐增强,促凋亡蛋白 Bax、 p53 随之下调, 抗凋亡蛋白 Bcl-2 则显著上调;

Objective:To investigate the effects of different concentrations of hydrogen peroxide on viability and the level of
apoptosis-related proteins in endothelial progenitor cells derived from human peripheral blood.Methods:Endothelial progenitor cells
were isolated from human peripheral blood by density gradient centrifugation method and differential velocity adherent method. After
passage, the third generation EPCs were treated by the final concentrations of 50 umol/L, 100 umol/L, 150 umol/L, 200umol/L, 300
umol/L and 400 umol/L H2O2 for 12 h, respectively; meanwhile, the control group was established. Cell viability was detected by CCK-8
assay. The expression changes of apoptosis-related proteins Bax, Bcl-2 and p53 in endothelial progenitor cells were assessed by Western
blot.Results:Exposure to low concentrations (≤ 150 umol/L) of hydrogen peroxide not only promoted EPCs viability, but also caused
decrease of proteins Bax, p53 expression and increases of the protein Bcl-2 expression. High concentrations (≥ 200 umol/L) of hydrogen
peroxide resulted in endothelial progenitor cell death; moreover, the level of proteins Bax, p53 expression was up-regulated while the level
of protein Bcl-2 expression was down-regulated as concentrations of hydrogen peroxide increased.Conclusion:Treated by different concentrations of hydrogen peroxide, endothelial progenitor cell viability and the level of apoptosis-related proteins Bax, Bcl-2 and p53 exhibited biphasic response. |
