目的:检测成肌细胞钙网蛋白(CRT)在循环拉伸应力刺激下的表达变化。方法:体外构建面颌部成肌细胞力学刺激模型。加
力组以0.5 赫兹的加载频率和10%细胞拉伸变形幅度对细胞进行加力培养1 h, 6 h, 12 h, 24 h,运用实时荧光定量PCR 技术跟
Western Blot技术分别检测在周期性张应力作用下成肌细胞CRT 在基因水平及蛋白水平的表达变化。结果:当对细胞加力6 h
后,CRT mRNA及蛋白表达量开始增多,加力12 h后CRT mRNA及蛋白表达量到达最多(P<0.01),加力0h 组与加力12 h组之
间差异有显著的统计学意义(P<0.01)。结论:持续的周期性张应力刺激下CRT mRNA 及蛋白表达增加。 |

Objective:To investigate the changes of Calcium protein (CRT) expression in myoblast under cyclic tensile stress
stimulus.Methods:The culture-tensile stimulate model of myoblast in vitro was established. The strength group cells were cultured for 1
h, 6 h, 12 h, 24 h under 0.5 hz loading frequency and 10%tensile deformation amplitude. PCR and Western Blot technique was used to
detect the changes of CRT in the genetic level and protein level under cyclic tensile stress.Results:After 6 h cyclic tensile stress, the
expression of CRT mRNA and protein began to increase, after 12 h cyclic tensile stress, the expression of CRT mRNA and protein reached
a maximum (P<0.01), the difference between 0 h group and 12 h has obvious statistical significance (P<0.01).Conclusion:The
expression of CRT mRNA and protein can increase under continuous tensile stress stimulation. |
