      摘要目的:通过检测各类型白血病骨髓中RASSF1A 基因启动子区甲基化水平,探讨其对白血病分型的临床检测意义。方法:抽 选93 例不同类型白血病患者(观察组)予以甲基化特异性PCP(MSP)方法进行骨髓RASSF1A 基因甲基化状态检测,研究不同类 型白血病甲基化状态差异,同期抽选93 例非白血病者为对照研究(对照组)。结果:观察组中有13 例(13.98%)检测到RASSF1A 基因甲基化,而对照组中RASSF1A 基因甲基化率为0%,比较差异显著(P<0.05)。不同类型白血病RASSF1A甲基化率比较:淋巴 系显著高于髓系(P<0.05),急性与慢性白血病比较差异无显著性(P>0.05)。结论:白血病骨髓中MSP 法检测存在RASSF1A甲基 化;而RASSF1A基因在淋巴系白血病中的甲基化概率明显增高,因此,对RASSF1A进行甲基化检测有可能作为白血病临床诊 断分型的生物学指标之一。

      ABSTRACT Objective:To investigate status of RASSF1A gene promoter methylation in bone marrow of patients with leukemia and the clinical significance. Methods:The selected 93 patients with different types of leukemia patients (observation group) by methylation specific PCP (MSP) method was taking for detection of methylation status of RASSF1A gene in bone marrow, Differences between patients with different types of leukemia state methyl, Over the same period 93 patients with non leukemia were selected as control study (the control group). Results:The observation group had 13 cases (13.98%) for detecting the methylation of RASSF1A gene, while in the control group, RASSF1A gene methylation rate was 0%, there was significant difference(P<0.05).Different types of leukemia RASSF1A methylation rate was significantly higher than that of comparison: lymphoid myeloid(P<0.05), acute and chronic leukemia had no significant difference (P>0.05). Conclusion:Detection of MSP by leukemic bone marrow in RASSF1A methylation; methylation probability RASSF1A gene in lymphocytic leukemia is obviously increased, therefore, the detection of RASSF1A methylation may be one of the biological index for clinical diagnosis of leukemia subtypes.
