目的:探讨三磷酸腺苷(ATP)对小鼠骨骼肌成纤维细胞的迁移作用及其可能机制。方法:细胞划痕实验检测1 uM、10 uM、
100 uMATP 对NOR-10 细胞愈合率的影响;细胞迁移小室实验检测空白对照组、100 uMATP 组、30 uMPPADS +100 uMATP
组、100 uMRB2+100 uMATP 组细胞的迁移率。结果:细胞划痕实验及迁移实验表明高浓度ATP 能够促进NOR-10 细胞的迁移
能力,100 uMATP 促细胞迁移能力最强(P<0.05),并且其促迁移作用能被30 滋MPPADS,100 uMRB2 所抑制(P <0.05),但
100uMRB2 的抑制作用更强(P <0.05)。结论:高浓度ATP(>10 uM)能够促进NOR-10 细胞的迁移能力,并且其促迁移可能通过
激活P2Y 受体作用大于P2X受体。 |

Objective:To investigate the effect of ATP on mouse skeletal muscle fibroblast cell migration and its possible mechanism.Methods: NOR-10 fibroblasts healing rate was evaluated under 1 uM, 10 M, 100 uMATP treatments by cell scratch assay; cell
migration rate was estimated by migration chamber in control group, 100 uMATP group, 30 uM PPADS+100 uM ATP group, and 100
uM RB2+ 100 uM ATP group.Results:cell scratch and migration chamber experiments indicate that high concentrations of ATP can
promote migration of NOR-10 fibroblast , and the capability is strongest by 100 uMATP treatment (P <0.05), and its role in promoting
the migration can be inhibited by either 30 uM PPADS or 100 uM RB2 (P <0.05), but the inhibitory effect is greater by 100 uM RB2
treatment (P <0.05).Conclusion:High concentrations of ATP(>10 uM)can promote cell migration of NOR-10 fibroblast, which is
probably through the activation of P2Y receptors more than P2X receptors. |
