      目的:观察山楂叶总黄酮对棕榈酸损伤的胰岛βTC3 细胞是否具有保护作用,并筛查出山楂叶总黄酮所起作用的有效浓度。 方法:以胰岛βTC3 细胞为研究对象,使用棕榈酸制作脂毒性模型,采用MTT 法观察山楂叶总黄酮是否具有保护作用,并进一步 筛查出有效浓度及时间;同时采用末端脱氧核苷酸转移酶介导的dUTP 原位切口末端标记(TUNEL)技术检测胰岛βTC3 细胞的 凋亡情况。结果:MTT 结果显示10-100 μg/ml 的山楂叶总黄酮对棕榈酸损伤的胰岛βTC3 细胞均有保护作用,其吸光度值明显比 棕榈酸组高(P<0.05),并且50 μg /ml 的山楂叶总黄酮与棕榈酸共同处理胰岛βTC3 细胞24 小时具有最好的保护作用;TUNEL 检测结果显示山楂叶总黄酮+ 棕榈酸组胰岛βTC3 细胞的凋亡率比棕榈酸组低(P<0.01)。结论:山楂叶总黄酮对脂毒性损伤的胰 岛βTC3 细胞具有保护作用,并在10-100μg/ml 浓度范围内成一定的剂量依赖效应。

      Objective: To obverse the effects of hawthorn leaves flavonoids on the apoptosis rate of the islet βTC3 cells induced by free fatty acids (FFA), and its effective concentration was also investigated simultaneously. Methods: The research object used were the islet βTC3 cells, and palmic acid was used to produce lipotoxicity model. The inhibitory actions of FFA on the proliferation of islet βTC3 cells were observed by using MTT assay and the effective concentration of hawthorn leaves flavonoids and treat time were investigated in details. The apoptosis rate of the cells was detected by TUNEL method. Results:The results obtained from MTT indicated that the hawthorn leaves flavonoids could effectively inhibit the lipotoxicity of palmic acid, and its absorbance is greater than that of palmic acid groups (P<0.05). On the other hand, the treatment of the 50 μg/ml hawthorn leaves flavonoids together with palmic acid to the islet βTC3 cells for 24h was demonstrated to be the most effective in cell protection. The results of TUNEL shown that the apoptosis rate of the hawthorn leaves flavonoids and palmic acid group is lower than that of the palmic acid group. Conclusion: Hawthorn leaves flavonoids could bring an effective protection for the islet βTC3 cells during the stimulation by free fatty acids, and this protection is dependent on the concentration of the hawthorn leaves flavonoids.
