法：免疫细胞化学和Western blot 方法检测使用5- 氮杂-2'- 脱氧胞苷处理细胞前后细胞内SOCS-2 表达水平。MTT 法检测单独使
用5- 氮杂-2'- 脱氧胞苷或顺铂及两药联合使用对CP70 细胞的抑制作用。结果：两药联合处理CP70 细胞后顺铂耐药逆转倍数为
1.34、1.63、2.34，联合处理组的细胞中SOCS-2 表达明显升高。结论：5- 氮杂-2'- 脱氧胞苷可以部分逆转CP70 细胞对顺铂的耐药
性，且此作用可能与增加细胞的SOCS-2 表达有关。 |

| Objective: To investigate the effect of 5-Aza-CdR on resensitizing cisplatin-resistant human ovarian carcinoma ceils
CP70 and discuss the related mechanism. Methods: Immunohistochemical analysis and Western blot were used to detect the expression
of SOCS-2 in different groups. MTT assay was used to assess the inhibitory effects of 5-Aza-CdR, cisplatin alone or a combination of
both on CP70 cells. Results: The resensitizing fold of cisplatin combined with 5-Aza-CdR was 1.34, 1.63, 2.34. The expression of
SOCS-2 had the most prominent decrease in CP70 cells in the combination group. Conclusion: 5-Aza-CdR can partlly resensitize cisplatin-
resistant CP70 cells, which might be related to the down-regulation of SOCS-2. |
