48h,检测其对成骨细胞增殖的作用;用碱性磷酸酶试剂盒检测ALP 活力。结果:在染铅48h 后,当铅浓度≥10μmol/L 时,细胞增殖
功能下降(P<0.05);加锌干预48h 后,铅+ 锌组细胞增殖功能均高于各自单独染铅组,其中铅(1μmol/L、10μmol/L)+ 锌(50μmol/L)
组、铅(10)+ 锌(100)组与对照组间的差异具有统计学意义(P<0.05)。铅干预48h 后,100μmol/L 铅组的ALP 活力显著下(P<0.05),给
予锌干预的铅锌联合染毒组,各组ALP 活力均有增加,其中铅(1μmol/L、10μmol/L)+ 锌(50μmol/L)组ALP 活力均高于对照组,而
铅(100μmol/L)+ 锌(50μmol/L)组ALP 活力低于对照组,差异均有统计学意义(P<0.05)。结论:铅对成骨细胞有毒性作用,影响其增
殖和分化功能;50μmol/L 锌在一定程度上可以拮抗铅对成骨细胞增殖和分化功能的损伤,且对ALP 活力的作用更显著,为铅中毒

Objective: To investigate the effects of lead and zinc on proliferation and differentiation of osteoblasts in newborn rats.
Methods: OsteobLasts were isolated from the calvaria of newborn rats and cultured in α-MEM medium. Lead and Zinc acetate with
different concentration was administrated and cultivated for 48h.The proliferation response to lead and zinc was determined by MTT
methods. The activity of alkaline phosphatase (ALP) was measured by alkaline phosphatase kit. Results: Treated with different levels of
lead (10 μmol/L and 100 μmol/L) for 48h, the cell proliferation decreased(P<0.05); Treated with zinc and lead for 48h, cell proliferation
was higher than their single lead-treated group; differences were of statistical significance (P<0.05) between the lead (1 μmol/L) plus zinc
(50μmol/L), lead(10μmol/L) plus zinc (50μmol/L), lead(10μmol/L) plus zinc (100μmol/L) and control group. The activity of ALP was
significantly decreased in group treated with a dose of 100 μmol/L lead for 48h (P<0.05). When treated with zinc and lead for 48h, the
activity of ALP was higher than their single lead-treated group; the ALP activity in groups of lead (1 μmol/L,10 μmol/L) plus zinc
(50μmol/L) were higher than the control group, while that in group of lead (100 μmol/L) plus zinc (50 μmol/L) was significantly lower
than the control group. All the difference was statistically significant (P<0.05). Conclusions: There were adverse effects of lead on
osteoblasts such as affecting their proliferation and differentiation. Zinc (50μmol/L) may have antagonistic joint action on lead in affecting
the proliferation and differentiation of osteoblasts. Moreover, it also has a notable role on the activity of ALP. So it provides some
proofs in preventing and curing lead poising bone diseases |
