RT-PCR 方法分析3 株人乳腺癌细胞和1 株正常乳腺上皮细胞中Plk1 基因mRNA 的表达水平。同时分析84 例乳腺癌及对应的
癌旁正常乳腺上皮组织中Plk1 mRNA 的表达水平。统计学分析Plk1 mRNA 表达水平与乳腺癌患者年龄、肿瘤大小、组织分化程
度、淋巴结转移状况、TNM 分期和雌激素受体(ER)等临床病理参数之间的关系,以及与预后之间的关系。结果:Plk1 基因mRNA
在乳腺癌细胞中的相对表达水平显著高于其在正常乳腺上皮细胞中的相对表达水平(P 值均小于<0.05)。另外,Plk1 mRNA 在乳
腺癌组织中平均表达水平(0.88±0.18)显著高于其在癌旁正常乳腺上皮组织中平均表达水平(0.22±0.10;P<0.01)。统计学分析结
果表明:Plk1 mRNA 表达水平和乳腺癌患者的淋巴结转移状况及TNM 分期密切相关(P=0.009 或0.007)。Kaplan-Meier 生存曲线
分析结果表明:高Plk1 mRNA 表达水平的乳腺癌患者的5 年无疾病进展率及总体生存率均显著低于低Plk1 mRNA 表达水平的
乳腺癌患者(P=0.0026 及0.0136)。COX 模型的多因素预后分析结果表明:Plk1 基因mRNA 表达水平是乳腺癌患者的一个独立的
预后因素(HR=4.764, 95%CI: 1.341~6.123, P=0.0025)。结论:Plk1 在乳腺癌组织呈现高表达水平,其mRNA 表达水平有望成为临

Objective: To determine the expression of Plk1 mRNA and evaluate its prognostic value in breast cancer. Methods: Semi-
quantitative RT-PCR assay was performed to detect the expression of Plk1 mRNA in three human breast cancer cell lines and one
normal breast mammary epithelial cell line. Additionally, the expression of Plk1 mRNA in 84 breast cancer and corresponding normal
breast epithelial tissues was also detected. Statistical analysis was performed to analyze the associations of Plk1 mRNA expression with
clinicopathological factors of breast cancer patients including year, tumor size, tissue differentiation, lymph node metastasis, TNM stage
and ER status. Finally, the association of Plk1 mRNA expression with prognosis of breast cancer patients was also analyzed. Results: The
relative expression level of Plk1 mRNA in human breast cancer cells was significantly higher than that in normal mammary epithelial cell
line (P<0.05). Additionally, the relative level of Plk1 mRNA expression in breast cancer tissues (0.88±0.18) was significantly higher
than that in corresponding normal breast epithelial tissues (0.22±0.10;P<0.01). Statistical analysis showed that the level of Plk1 mRNA
expression was significantly correlated with lymph node metastasis and TNM stage of breast cancer patients (P=0.009 and 0.007, respectively).
Kaplan-Meier curves showed that the 5-year progression-free or overall survival rate of breast cancer patients with high Plk1 mRNA
expression was significantly lower than that of breast cancer patients with low Plk1 mRNA expression (P=0.0026 or 0.0136). Furthermore,
Multivariate survival analysis using Cox's regression model showed that the status of Plk1 mRNA expression was an independent
prognostic factor for breast cancer patients (HR=4.764, 95% CI: 1.341~6.123, P=0.0025). Conclusion: Plk1 mRNA is highly expressed in
human breast cancer tissues, and might be an important prognostic factor for breast cancer patients. |
