目的: 研究骨髓间充质干细胞(MSC) 对大鼠脑缺血再灌注损伤的治疗机制。方法: 20 只Wistar 大鼠随机分为对照组
和MSC 治疗组。应用GFP 阳性MSC, 再灌注1d 后经尾静脉注射MSC( 1 ?? 106) , 对照组则注射PBS。采用线栓法建立脑缺血再灌
注模型。术后每天由双盲于试验组的研究人员应用爬杆计分法评定大鼠神经功能。缺血2h 再灌注8d 取脑组织, 用免疫组织化
学方法检测脑组织中bFGF 的表达。结果: MSC 治疗组大鼠的神经功能缺损评分明显低于手术组和对照组( P< 0. 05 ) 。MSC 治
疗组缺血侧缺血周边区脑组织中观察到GFP 阳性与bFGF 免疫组化染色阳性细胞。结论: 经尾静脉给予的MSC 可促进脑缺血再
灌注大鼠的运动功能恢复; bFGF 表达升高, 可能是MSC 脑保护作用机制之一。 |

Objective: To investigate the mechanism of mesenchymal stem cells (MSCs) treating cerebral Ischemia- reperfusion in??
jury of wistar rats. Methods: 20 Wistar rats were randomly divided into control group, MSC- treated group. One day after the reperfusion, 1
x106 MSC were injected via tail vein and the same dose of the PBS was given to the control group. Cerebral ischemia reperfusion model was estab??
lished with the suture method. 8 days?? reperfusion was given to the rats, after 2 hours of middle cerebral artery occlusion (MCAO) , whose cere??
bral tissues were obtained. The neurological deficits of the rats were evaluated by the blind investigator of the research every day with the modified
neurolog ical Severity Score Points. The MSC were detected by Laser Scanning ConfocalMicroscopy and the basic fibroblast growth factor ( bFGF)
expression was detected by immuno- histochemistry. Results: The score of neurological impairment in the MSC- treated group was significantly
lower than that in the control group ( P< 0. 05) , with GFP and bFGF immunochemical stain positive cells around the ischemic brain. Conclu??
sion: MSC given via tail vein can promote motor function recovery of cerebral Ischemia reperfusion rat. The increase of expression may be one of
the MSC?? s neuro- protective mechanisms. |
