The optimized freeze- drying cryoprotectant of Acidithiobacillus ferrooxidans was studied by orthogonal experiment, in
which glycerol, trehalose, sucrose and bovine serum albumin ( BSA) were regarded as factors. The results of Direct- viewing analysis, the factor
index analysis and variance analysis indicated that in freeze- drying cryoprotectant, which is composed of glycerol, trehalose, sucrose and BSA,
the gradation of influencing survival rate is in turn as follows: glycerol > trehalose > sucrose > BSA. The optimized combination of freezedrying
cryoprotectant consists of 5%( w/ v) glycerol, 15%trehalose, 18%sucrose and 10% BSA. , in which, the survival rate of Acidithiobacil??
lus ferrooxidans could reach 94%. |
